Description

Anti-DLL3 VHHs were isolated from humanized yeast-displayed VHH library, expressed in HEK293 with Fc of human IgG1 [1-2].

Flow Cytometry

Methods: 

1. VHH displayed yeast cells were incubated with Human DLL3 and anti-V5 at room temperature  for 30 minutes. Wash twice. 

2. Incubated with AF647 conjugated Streptavidin  and AF488 Conjugated Anti-Mouse IgG at room temperature  for 30 minutes. Wash twice. 

3. Flow cytometry analysis.  The forward scatter (FSC) signal is for expression and display level, and the side scatter (SSC) signal is for binding affinity.

Results:

Enzyme-linked immunosorbent assay (ELISA)

Methods:

1. Coat the plate with 100ul/well of 500ng/ml of human DLL3 overnight at 4°C. Wash the plate with PBST for 3X.

2. Block the plate  with 200ul/well of 3% BSA/PBS for 1 hour at RT. Aspirate the blocking buffer.

3. Add 100ul/well of serial diluted VHHs to plates. Incubate at RT for 1 h with gentle shaking, and wash plates with PBST for 3X.

4. Add 1:1000 dilution of anti-hIgG Fc-HRP and incubate at RT for 1 h with gentle shaking. Wash the plate with PBST for 3X.

5. Add 100ul/well of substrates and read the plate within 5 min.
